King's Highway 140 runs north-south, roughly following the eastern side of the Welland Canal, connecting Welland to Port Colborne.
 
Northern Terminus: Intersection of East Main Street/Niagara Region 27 in Welland.
 
Southern Terminus: Junction of King's Highway 3/Niagara Region 3 in Port Colborne.

Approaching 140 from the Main Street Tunnel. This stretch of roadway through the tunnel linking 406 to 140 is maintained by MTO and carries the unsigned designation of Highway 7146.

To the east, Netherby Road links to the eastern terminus of King's Highway 58A and the Townline Tunnel under the Welland Canal. Unlike the Thorold Tunnel and the Main Street Tunnel, the Townline Tunnel has only one bank dedicated to motor vehicles, as the other carries train traffic.
